I guess that could be done by placing an mp3 in a predefined location
(.rockbox/startup.mp3 for example).

The patch that exists (FS#6097 -
http://www.rockbox.org/tracker/task/6097) simply adds a string to the
voice file for both messages. So that would be more automated, but less
configurable. A good reason to do it this way is that it would work on a
user's very first boot with voicing enabled, so you'd get a confirmation
that Rockbox is running and voice is working.

A way to do both would be to check for the MP3 clip, and if that doesn't
exist, use the voice-string.
